There should be an option in your control panel. Instructions below are for ATI cards.
Right click -> Properties -> Display Tab -> Advanced -> 3D Tab -> Check the custom box -> click on custom -> move "wait for vertical sync" slider right -> click apply and then ok
If you only want V-Sync for that game, you can save a specific profile for that game. That way you can change settings like V-Synce, AA and AF for different games fairly fast.
