This is what I think City 17 is, based on the information from all the HL² videos.
the number 17 is just it's designation; cities are turned into containment camps to keep humans in order and to make them work. In the latest e3 video, the people are talking about how the trains leave full but come back empty. The people are being taken to work camps or maybe death camps... they're stripping the planet of natural resources. The Combine probably has done this before, and is just moving on to another planet, enslaving the population, using them to cut down trees and mine and stuff, then kill them all and move on to the next world.
This is what I think the combine may be:
Humans who gave up some of their humanity to be melded with alien DNA (like the Wraeththu, if anyone's read those books by Storm Constantine) because they saw it as the only way to survive, or the next step in evolution, or whatever. That accounts for the humanoid bodies and the name Combine. the aliens might be the enemies of the G-man or something... the reason the combine are so ruthless and mean (we see the bloody chairs in the interrogation rooms or whatever they are in the video) is because the alien DNA altered the human's emotions.
I dunno, just some of my thoughts.
