Just throw out some examples here of characters in games that are pure awesome. I'll add more later, I'm supposed to be studying. :D

Myron (Fallout 2, Interplay, 1998) - from his initial starting conversation where he possibly mistakes you for a whore, to his egotistical spouting of scientific bullshit, to the fact that he invented the most addictive designer drug on the planet, Myron is a barrel of laughs from point a (where you meet him) to point b (Where you shoot him, because then you steal his stuff.) Or you could have him join the party. It's probably worth it for his dialogue. (although he really blows at combat).

The Entire Cast of Psychonauts (Psychonauts, Doublefine, 2005) - "My name..." ("Starts with a 'D'!") "...is Razputin. But everybody calls me... Raz." This game is genius and every single character in it is genius. Linda the Lungfish! Mr Pokeylope! Sasha Nein and Mia Vordello! Oleander! Lili Zanatto and Raz! Some of the most fun characters ever invented.
 
Leon Scott Kennedy(Resident Evil,2,4)-It's his first day as a police officer in Racoon City,and,upon arrival,discovers a bloody body in the road,suddenly,zombies begin to attack(epic cutscene)In Resident Evil 4,your called to rescue the Presidents daughter,and you end up saving the world

Albert Wesker(most Resident Evil games)-comparable to the Gman,except,you do see him in life and play as him in one

Bobby Sawer(Juiced 2)-IDK,he won my camaro z28 in a pink slip race,and is overall the most fun to challenge

Sephiroth (FF7)- the video game worlds ultimate bad ass evil villain
Mario - obvious reasons
Auron (FF10) - the worlds ultimate bad ass good guy

Manny Calavera is one of my tops. I really enjoyed playing as him and I could, in some strange way, identify myself with him.

Max Payne & Mona Sax (Max Payne 1 & 2) - These incredibly noir movie-like games were the pinaccle of gameplay in their time, but their story was also one of the best I've ever seen. Max Payne is perfectly fleshed out while Mona Sax remains interestingly unknown in both series. Both endings of Max Payne 2 almost made me cry (teary-eyed ;().
 
Stick to the format, gents.

Dr. Breen
(Half-life 2, Valve, 2004): One of the most well-written, well-acted villains in the history of gaming. An intriguing character from the off, Breen gradually goes from cold, logical-minded collaborator to a desperate, arrogant man seeing ten-years of hard work being flushed down the crapper (“What is it exactly that you have created? Can you name even one thing? I thought not.”). Bonus points for his science driven, intelligent speeches on the “future of the species”.

Tommy Vercetti (Grand Theft Auto: Vice City, Rockstar, 2002): A complete and utter badass, and he was an absolute thrill to play as. Words cannot state how fantastic Tommy Vercetti is. He's just that ****ing cool.

There's more, but I can't be assed to list them.

I agree, but Kefka isn't "the video game worlds ultimate bad ass evil villain." He's extremely evil, but "bad ass?" No.

VAL (Virtual Artificial Lifeform) (Startopia) - One of the wittiest and sarcastic AI I've ever witnessed.

Jack (Vampire The Masquerade: Bloodlines, Troika, 2004) Oh yeah. VTM had the best voice-acting ever, period, and Smiling Jack was one of the most awesome non-player characters ever. I thought he'd be a simple tutorial dude at first, but he was just so much more awesome than just a tutorial teacher.

Yeah I guess so. Noticed it myself after I posted.


But I can't really think of any favorite human characters. There's the Milkman from Psychonauts, but he's been mentioned (generally).
 
G-Man (Half-Life Series) - Ultimate mystery, great enigma.

Liquid Snake (Metal Gear Solid) - Awesomeness on many levels, cool, British, ingenious and spectacular dark twin. Also advocate of the selfish gene theory.

JC Denton
(Deus Ex Series) - He was great in first Deus Ex but after his merge with Helios he become truly awesome. Behold your transhuman God!

HK 47 - has one of the greatest personalities ever.

Sam and Max - They are just too funny.

Augustus Cole (GoW) - Sure, he's a stereotype, but his one liners and overall attitude adds more to an already awesome game.

Glados - Fiendishly clever, and has a tremendous amount of humour to boot.
 
G-man: Great lines, mysterious, and sinister.

Dr. Magnusson: The fact that he's a pompous bastard, with little feelings. Why else would he be my avatar?

Alyx: By far the most humanised character I have ever seen in all my years as a gamer.

Big Boss(Metal Gear series)- His salute at the end of Mgs3 at the Arlington National Cemetery=epic win.
The Boss(MGS3) - the most badass female character in gaming imo, when she reveals her motives at the end of mgs3 was amazing, 'loyal to the end'.
Solid Snake(Metal Gear Series) - the ultimate gaming hero?
Agent 47 - ''This room. This bullet. There's a bullet for everyone. And a time. And a place. Yes... maybe this is how it has to be.

Inspector, you've obviously learned too much about me. I can't have that. Even in my death'...

All classes / Team Fortress 2 - can't beat these guys for offbeat violence and undying commody. Their artistic and game-mechanical functions are perfectly melded, as their hilarity helps you play the game by distinguishing them. A real pinacle of design.

GLaDOS / Portal - Funny. Evil. Sympathetic. Informative. What more do you need?

Borden / Hostile Waters - All the characters in this game were good - you had to place the recorded personalities of dead soldiers into your war vehicles, and they were all different and would constantly bicker with each other. Borden's a pretty random pic, but she's my favourite for her charmingly freaked-out reaction to being brought back to life as a computer program. Tom Baker should get a mention for his narration.

Breen / Half-Life 2 - cf. Samon

Zyzyx the imp / Sacrifice - best helper character in an RTS ever. "Your creatures are dying!"
